I have been trying to estimate the carbohydrate present in Oscillatoria sp., but with erroneous results.
Procedure Followed: Heating the algal sample for 3hrs in HCl - Adding Anthrone reagent - OD measurement after cooling.
Can anyone please provide me with another protocol for assay, excluding FT-IR (Since I have no provision for it).

I use the Dubois method (1956) which needs H2SO4 solution+ 5% phenol sol. + incubation at 90 C + OD@490 nm. Calibration curves may be done with Glucose or Sucrose or Starch. Final result comes in Glucose equivalents.
