I started with 3mL human CSF to isolate Exosome by using ExoQuick-TC Ultra kit. After mixing and incubation with 0.6mL ExoQuick-TC O.N. at 4C, 3000Xg centrifugation didn't produce any visible pellet. Is this normal? Can you see the pellet?
Shamsher S. Kanwar Thanks for recommending the paper! In Figure 3 of this paper, the centrifugation speed for ExoQuick/Plasma mixture is only 1,500g, even half speed than 3,000g I used.
In fact, I can isolate exosome from Plasma normally. As for CSF, I used ExoQuick-TC Ultra kit and followed the protocol in the manual, but couldn't see a pellet after centrifugation of ExoQuick-TC/CSF mixture.
I asked this question, because in the manual of the kit (ExoQuick-TC Ultra) I used which is used for isolating exosomes from tissue culture medium, it is said that "after centrifugation, the EVs may appear as a beige or white pellet at the bottom of the tube".
I did see a big pellet when I isolated exosomes from plasma using ExoQuick kit.
