I work at Camosun college. I am the technologist that makes up PCR mix- Master mix, water, PV 92 forward and PV 92 reverse primers for a PV92 lab. In some cases, I may have extra amount of primers. Can I refreeze them and use in a later lab (3 months later)?
Yes primers are quite hardy and will withstand freezing many times. Even so when you thaw this stock oligo it would be best to aliquot a few samples so that the master stock is not thawed too many times.
Depends on what the primer is dissolved in/diluted with. If dissolved in TE then they will be fine but if dissolved in water there is be slow degradation over time but 3 months is not that long.
I have used my primers after 6 months also. Yes, the primer diluent makes much difference. I used autoclaved nuclease free water for the dilution of working primer concentration and TE for the stock. Still if you doubt, run your individual primer in 3% agarose gel for not more than 10 minutes. Here, if you don't get the smear then your primers are fine. Hope this helps you.
Yes, but I would make aliquots that are sufficient to cover your needs and minimize the refreezing of your stock primer. I would also consider synthesizing new primers after a few years or multiple freeze-thaw cycles.
