I need to evaluate MIC activity of some plant extracts on C. difficile. Can anyone suggest a medium for C. difficile? Can something other than brain heart infusion be used? How long should cultivation least?
Usually for anaerobic bacteria to perform E-tests for MIC determination recommended medium is Brucella agar with blood. This is the best choise. If you are afraid to add blood, you can use Brucella agar or try Columbia agar.
Depends on what criteria you want to use. CLSI uses BBA indeed, as Gayane Martirosian indicated. If you want to use microbroth dilution rather than agar dilution, you can use BHI as well. Be aware that some antimicrobials have a requirement for divalent ions, and this may have to be added to the medium.
