I am using microfluidic device for cell culture experiments. I need to fix cells with PFA for ICC. I was wandering if we can remove the cells(clean the device) after ICC so that the device can be reused.
As a physiologist, I do not recommend using fixatives in equipment or tools that are used with live cells. Fixatives, such as PFA, are hard to wash and small residues can stuck to the equipment can kill cells for a long time. Furthermore fixative may cause damage to tubing. Therefore I would not recommend it at all, especially if you are sharing the facility with others, as you may effect their experiments.
Do you construct your microfluidic devices so that the PDMS is permanently bonded to the glass substrate on which the cells are growing? If that is the case, I agree with Refik and suggest that you do not plan on reusing the device.
Thank you Refik and Jill for your input. @Jill, Yes I am fabricating the device in the same way. Can you suggest any other fixation method which I can use for ICC. Can the PDMS device be reused if we are not going for ICC in the device.
