Hello everyone,

I would like to know whether we can use the vibratome to section zebrafish larvae (8-9 days post fertilization) for histology. We have a Leica VT1200S vibratome in our lab. According to the instrument specs we can cut 50 micro meter sections after embedding samples in agarose. But the problem is I need to cut 4-5 micrometer thin sections using this vibratome. Previously, we used the conventional microtome to cut 4-5 micrometer sections after embedding with paraffin.

1. Is it necessary to do the ethanol dehydration process before vibratome sectioning?

2.Do we need to keep the samples in xylene before vibratome sectioning?
