I am trying to study the matrix effect by standard addition. From my reading, I found a standard addition method that is used to study ion suppression and enhancement. Aliquots of the case sample are spiked with different concentrations to build up a linear regression. Dividing the intercept with slop gives the concentration of the target analyte in the case sample.

Thank you for your comment. What I know is that the optimal enzyme activity in 1 ml is between 7000-8000 units. So based on it if my urine sample volume is 100 ul. Therefore, I need 70-80 units. so if the enzyme bottle is 50 kU/ml so we need 1,4 ul of the enzyme. But I am doubtful in this way as the volume of the enzyme is very low and I do not know how much I need a buffer volume. The link contains a manufacture recommendation, I hope it would be helpful.
