I am working on steroid estrogens concentration in lettuce plant. Can we measure estrogens from green extract of plants on GC/MS? I am afraid, if I use dark green sample may cause some problem in GC/MS . I have tried many times, SPE cartridges but recovery percentage is very low. Which is most suitable and cost effective method for color removal? Please share your Ideas and experiences.

I think you should try to reduce color of your sample by passing your sample through column, and not to worry about GC-MS, it will help to find area peak (%) of estrogen in your sample.

Just for the initial assessments of the Component behaviours, I will like to spend some time on the TLC of the Extracts, see the solvent system efficiency to separate the components , then go for GLC Column separation campaign. This is so that we dont damage the Expensive GLC Column. Off course , I will never use the unpurified extract directly on the GLC Machines. Whatever technique we use for extract purification , we must go through this exercise first to have a slightly purified extract for the Columns. We may get into, esp. when we are the beginners, new problems of wasting time & energy & still getting no where.
Is there a relationship between benzene green colour and steroids? I used a GC-MS analysis of some plants and showed a high percentage of steroids, including steroids estrogen regardless of the colour of the plant extract. You may need to change the extraction method or the type of extract you are using. The type of column used and all that is mentioned must be consistent in terms of polarity
