I tried to understand the Nitrogen Use Efficiency of the newly released taro varieties using two factors, taro varieties as the main plot at 4 levels and fertilizer rates as the sub-plot. The design to be used is the split-plot. The third factor that confused me was the destructive sampling at 40-days intervals.
Could the destructive sampling be the third factor or just a standard sampling approach?
You can use your split plot design with taro varieties as the main plots and 4 levels of Nitrogen as the sub plot. I would suggest using 0 40 80 and 160 as your nitrogen levels.
You will want to have 4 or more plots in the subplots to discern differences based on degrees of freedom. Genergy have a 6 row plot harviesting the center two rows for final yield and sampling the border rows for your time samples.
Your sampling is not factor for the experiment but rather just data for prescribed analysis based on the plot design.
Think about the number of plants you need and do all you can to have uniformity in the plots to allow the treatment factor differences to be expressed.

Charlie Suruban , What is the objective of the destructive time interval? If you want the trend of N efficiency of the varieties released over time, I do not think that time interval could be an independent factor. It will be complicated as the number of factors increase and you will have several confounding effects. Please use the two factors and look them what would happen and their effect over time!
Destructive sampling couldn't be a factor to be compared but a procedure to collect, classify, summarise and interpret the data for finding experimental results....all those sampling will be considered as destructive sampling where whole plant sample has to be taken into consideration rather than counting any of the growth attributes (like number of leaves, number of plants, number of panicles, number of branches etc) without damaging plants...One factor (main plots) with 3 levels viz. different cultivars while another factor with 4 levels (sub plots) viz. fertilizers rate ..here in split plot design, experimenter is interested to observe interection effect and sub plot treatment effects more precisely rather than main plot treatments....there would be 12 treatment combinations means 12 plots and if replicated 3 times then 36 plots of the same size. Border and sampling areas from where plant samples for observing many parameters during different time of interval (days after planting DAP) have to be collected from each 36 plots for statistical analysis...
