I have been trying to extract essential oil from leaves and flower calyx portions of marigold but not getting the optimum/ expected GCMS result i.e. the compounds i get are very few and also doesnt match with most or any of the articles availble (same article conditions were used for GCMS)
Very few peaks I am getting though the oil extracted has all the smelling properties.....
Till now for calyx portion I followed hydrodistillation (my meaning," Put the fresh sample finely chopped in a round bottom flask, pour approximate amount of D.water and extract using Clavenger's apparatus at 60oC for 2-3 hrs...the steam obtained is condensed (has the oil smell) and collected in amber bottles). Later they are collected and refluxed with Hexane/ Petroleum Ether (but no combinations) in a seperating funnel and the solvent layer is collected and taken for Rotatory evaporator for concentrating...(3-5 ml is the final volume generally I get for analysis)..
Result: the peaks didnt match and didnt obtained many compounds (getting almost 10-15 compounds) against expectation of 30-50 compounds
For leaf also I followed the same but without any appropriate result...
In other side, i tried directly with Hexane as the solvent instead of DW as extraction solvent in clavengers.....did the concentation...still no result in GCMS.....
Friends i am not understanding the mistake I am making....but what ever the samples after extraction and concentration do smell like the original plant material in all the cases.....
