I want to use NaN3 as a preserver for non-covalent BSA aggregates. In the future, I will want to use the aggregates to evaluate the innate immune response in another laboratory.
Do you know, if NaN3 at 0.02% can interfere in that experiment?
Can you recommend another preservative for 6 months of storage?
NaN3 is highly toxic. It is a ATPase inhibitor? (check this out). Never use this as a preservative in proteins, if proteins are to be used in cell culture or anything alive. It is OK for structural biology purposes in protein preparations. But certain enzyme reactions do get "poisoned" by NaN3. Please be careful.
Glycerol is only thing that comes to my mind. But you need to dialyze it out before use. You want to use the protein in a "innate immune response experiment", so better think hard about all the ingredients in your protein.
Hi Pilar, azide is good preservation agent for proteins kept at +4C for ~1 week, But for long term storage of proteins, you should keep them at -80C with azide. They will be stable for years.
You can get rid of the azide by running your BSA on a small desating column.
You probably don't want to freeze or freeze-dry your sample because those treatments might alter the non-covalent aggregates. Perhaps you could use thimerosal as a preservative.
