I have a purified protein and I am wondering if it is possible for the protein to migrate from top chamber to lower chamber in the absence of any cells using the Boyden chamber assay. I've attempted a few trials and it seems that it is taking over 24 hrs for the protein to be detected in the lower chamber. So I am wondering if I can use this assay to trace proteins only or do I need to have cells in there?
