Anyone how is an expert in UV spectrophotometer please guide me can I use seed sampled digested for atomic absorption spectrophotometer (in HNO3 and H2O2) for determination total protein contents.?
I doubt you will see any proteins left after digesting in acid and peroxide. The two solutions are intended to break down organic materials (which includes proteins) into their constituent parts, solubilizing the metals and other elements.
You won't even be able to measure the N content accurately as you used nitric acid.
There is used only mass spectrometry for such analysis. The quantitative analysis is easy and absolute, operating up to fmol level of analytical concentration. It can be carried out by the method shown in reference [1]. The employment of UV, or any other spectroscopic method does not provide reliable results due to error contribution obtained as a result from the mathematical processing of the spectroscopic patterns. A relatively comprehensive analysis in the latter contenxt can be shown in references [2,3]. Furthermore, it is well-known fact that all optical spectroscopic methods operate at high LODs and LOQs.
