Hi,
I want to preserve my field caught dengue mosquitoes in RNAlater solution until RNA extraction later. Before that, I want to identify and sex them. Since identification of live mosquitoes would damage their body parts, i am going to cold anesthetize them at 4 celcius for about 10 minutes and do the sorting. Once it is completed I am going to preserve them in RNAlater and put them in the freezer. I want to know if it is ok to add the cold mosquito to RNAlater. Would it do any degradation to the RNA of the mosquito?
if you have any better option, please suggest.

The way you want to perform on your mosquitoes is OK, but I prefer to identify them in anesthetized condition, then preserve it in -80 up to 1 year until carrying out the RNA extraction. If you do not have access to -80, you can try RNA later.

Thanks a lot. Do you have any idea whether using ether would affect the RNA of the mosquito?
The right way to to preserve RNA using RNA later is to keep the mosquito in RNAlater at 4degree celcius for 24h (during this time the mosquito will be fully impregnated) then you can put it in -20

Do you use home made RNA later or from the company (?)
I want to keep my insect samples in RNA later for next generation studies (NGS), but not sure which company has suitable RNA later (off course not a expensive one) for this purpose
