I am looking to attach "folic acid" to avidin. What I have in mind is to use "carbodiimide". firstly, FA was dissolved in NaOH 1M with carbodiimide for 2H, and then avidin was added into the above solution. Can I conjugate folic acid on avidin with procedure? or maybe I must added something buffer?

As long as you are trying to join these entities through the amide linkage (or ester formation based on availability of suitable groups, i.e., mainly OH, CO2H) you could safely use the carbodiimides. The problem would be the byproduct removal and purification. Other options for amide formations are available, check in consonant with your structure and the groups participating in the reaction. if you can put a question here, can go to at least google to chek on the reagent and procedure easily!
Avidin is a protein. I would be concerned about the destructive effect on avidin of adding it to 1M NaOH. Try to use conditions closer to a neutral pH. Coupling reactions are often done in 0.1 M sodium bicarbonate.
