Molecular oxygen can quench luminescence signal of fluorophores. I want to know if I can attribute this phenomenon only to singlet oxygen generation in a organic media.
I plan to perform experiments with increasing oxygen concentration and calculate the quenching rate constant to make this relation
Yes and No. Firstly, how do you plan on increasing oxygen concentration? Yes to your question... if only you are sure the solvent and/or the oxygen generating pathway (reagents or so) does not interact with the fluorophore.
Is your fluorophore organic based (dyes etc)? If yes, bear in mind that nature of solvents (polarity) affect their optical properties.
So, you need to rule out these to follow through with your intended experiment. Good luck.
There is also a minimum energy of activation for singlet oxygen generation. Off the top of my head, this corresponds to ~600nm excitation.
To generate singlet oxygen, which is in a triplet state in the ground state, you need special organic molecules called photosensitizers, which have high ability for intersistem crossing to the triplet state (as a rule molecules having high atoms in their syructure). In the triplet state such molecules can interact with triplet oxygen via nonradiative excitation energy transfer that causes their nonradiative fluorescence quenching and singlet oxygen production. In other cases you cannot atribute fluorophore (what kind ??) quenching to singlet oxygen. There are a liot of reasons for this process: consentration, solvents polarity, etc..
