Someone already used petrifilm AC supplemented with MRS broth for enumeration of lactic acid bacteria? I have questions about this alternative method.
Another questions is, have you already used candle-jar technique? I would like to understand if the ambient is considered anerobiosis or microaerophilia.
I haven't added MRS broth to the Petrifilm AC before. You can purchase Petrifilms specifically for Lactic Acid Bacteria though. They might be more useful for you. MRS is selective for LAB but if you are using a pure culture for your studies then the bacteria will also grow on the Petrifilm AC.
The candle jars are an old school technique for growing anaerobic bacteria. If you don't have access to an anaerobic chamber/incubator or anaerobic sachets, then it is a cheap alternative for culturing anaerobic bacteria. I'm not sure how well it works for microaerobic bacteria. Give it a try though.
