The viral DNAs are sensed by IFI16 and induces caspase-1 activation and pyroptosis. Is it possible that caspase can target and cleave IFI16 and remove PYD domain?
In our hands, we have not detected proteolytic cleavage of IFI16 protein using various antibodies that were target against various domains. However, I would not rule out the possibility that under certain conditions the IFI16 protein is cleaved by proteases (caspases) to remove the PYD. Assuming that the protein is cleaved, the role of N-terminal truncated IFI16 protein in the regulation of innate immune responses (in particular, in the regulation of AIM2 inflammasome) would be different.
