I need to determine nitrate level in biological sample for which i am using griess reagent manual colorimetric method. i prepared standard using NaNO2 (1-100 micromolar conc.) and it was performed smoothly. Now i am facing problem when i am doing this in biological sample including plasma, tissue homogenates. i did deproteinization of sample using 10 % TCA. i got some literture that acidic condition can do loss of NO3 in sample so ZnSO4 /NaOH was suggested butit does not mentioned that how much conc. of znso4 or NaOH should be use. can anone provides me the good protocol ?
