I want to isolate the pure culture of green sulfur bacteria in petri dish. I am using the CP medium(ref- Frigaard et al., 2001, Chromosomal Gene Inactivation in the Green Sulfur Bacterium Chlorobium tepidum by Natural Transformation ) for isolation of GSB in agar. But i am unable to get green colonies. Whenever i am doing isolation i got white colonies only. So anyone suggest the procedure for isolation of GSB in petri dish and how will be the GSB colonies look like

Thank you for your reply. Yes i have kept my plates in anaerobic chamber only under normal light condition(tube light)

Kindly use the following anaerobic chamber for the anaerobic conditions. As it will provide the desired anaerobic condition. Hope this might help you
