I want to quantify the proanthocyanidin content in my plant extracts using colorimetric method or spectrophotometry. Can anyone suggest me a suitable protocol for the same?
4-(dimethylamino)cinnamalde aldehyde (DMAC) method has great potential as a rapid analytical method, DMAC is an aromatic aldehyde that reacts with flavan-3-ols and c-PAC to form a green chromophore with maximum absorbance at approximately 640nm.
3 mL of 4% (w/v in methanol) of vanillin should be added to 0.5 mL of diluted sample. 1.5 mL concentrated HCl should then be added to the mixture and left at room temp for 15 min before measurement of the absorbance at 500 nm. Catechin can be used as the standard for a calibration curve and the results can be expressed as mg of catechin equivalents per gram of sample (mg CE/g).
