What we want to do is target MRM experiment. We purified the target protein and did LCMS to identify those peptides from our target. Right now, we want to do MRM type of experiment to generate quantitative information about modifications on certain peptides. Any suggestion will be greatly appreciated.
You can't. Not with a G2. Not in the common MRM sense. You can't generate MRM peaks. I've been told that things have changed with new G2-Si, with an MRM feature added to Masslynx. All you can do with a G2 is to extract the fragment ion(s) current from the HE trace and use the corresponding peak area(s). It worked for me. Best
Many thanks Andrea! You are totally right. We got similar information from Waters. We can not do the common MRM on the G2. We did similar thing to extract the fragment ions current from the HE trace during MSE experiment. MRM may be a wrong name here. But we really want to do is kind of SRM. We only want to select certain peptide peaks to do the quantitative analysis on the G2.
