I hereby would like to know that whether the samples for Biological Oxygen Demand(BOD) test needs to be fixed prior to taking the readings if the Dissolved Oxygen(DO) readings are to be taken with a probe as in Winkler's method?
If you measuring DO by using Probe then it's better to determine your Initial DO on site and if you fix your DO as you doing in Wrinkler Method then it will not possible to measure DO by using Probe and it will be done only by titration method bcoz your all available DO were present in fixed form not in loose.
If you refer to the Standard Methods for the Examination of Water and Wastewater, then in the beginning you Will find that Do,pH, temp. must be measured instaneously at the moment of taking the sample. So a portable Do probe is a prerequisite. Best regards.
However, please keep in mind that for the BOD test, the field DO value is not needed. Instead, you want to know the DO (usually after bringing it to 20 C and aerating the dilution water or even the sample to get it above 8 mg/L) immediately before sealing the BOD bottle for the 5 day incubation period.
