I have been trying to find where I can order Agrobacterium tumefaciens GV3850 strain as my transformation is less efficient with LBA404 and EC58 strain that I have. Didn't find any source from where I can get Agrobacterium tumefaciens GV3850 strain. Any suggestions?? Thanks.
When we have a problem with transformation efficiency we use the Agrobacterium AGL1 strain, it's actually much better than any of the GV strains we tried you can orderit from the ATCC collection (www.ATCC.org)

Isabelle Malcuit thanks. but i am searching GV3850 because the vector I am using for construct had been proved for efficiency with GV3850. If i do not find this, i will go with your suggestions.
Which vector are you using? AGL1 works well with a range of vectors including Pgreen and pBIN type of vectors

unfortunately, I don't use this particular strain.
But, in my experience it is not so much the strain/vector combination having a strong impact on transformation efficiency as the host/strain combination. In my hands, for some species a "high" virulence strain with a certain vector would result in very few surviving transplants, while a supposedly low virulence strain with the same vector would work fine. On the other hand, the same two combos gave me the opposite result with a different plant species. So, I figure it's more about host range of the Agrobacterium strain. I would try to figure out which strain gives you the best transformation efficiency for your target species with some easy to detect model gene (GUS, GFP, antibiotic resistance) and then see whether it works for your vector as well. That may take some time, but it safes some frustration I guess :-)
