I am trying to plot the standard curve for NO scavenging assay with Ascorbic assay. I need this to test the NO scavenging ability of the plant extracts I have prepared. But the results are so confusing. They are impossible to reproduce and most of the time the control OD is less than the sample OD. So all the values are negative. Few times, the inhibition percentage linearly decreased with the increasing concentration. I have tried many different ways to run this assay. Can anyone please help me find the mistake?

This shouldn't be if you have had appropriate control and blank. A blank for the different concentrations of the drug itself is also necessary as the OD/colour of the drug preparation could also affect your result.
