Disc diffusion method
just soak the disc with your plant extract and place on inoculated plate.
dilution on microplates should be the best way. But, you should first of all know the the characteristics of the microbial strain (growth temperature, the number of hours to incubate). have a look on that paper http://www.biomedcentral.com/1472-6882/13/26
CLSI method is the best when your plant extract diluent does not affect the growth of microorganisms. If the diluent may inhibit or stimulate the growth of microorganisms would recommend disc diffusion method but using sterile paper disks impregnated with extract in several dilutions and dried. MIC can then be calculated from the transformed equation of linear regression.
