In my hands, there is no good commercial antibody against the 5-HT1A receptor. All the antibodies I have tested gave background in region not known to express the receptor (that can be identified with specific labelled ligand).

In situ hybridization identifies clearly the soma expressing the receptor, See the ref:

Have you tried to use the 24504 Serotonin receptor 1A (HTR1A) antibody of Acris Antibodies?

Thank you all for the very valuable and constructive comments and suggestions. I have tried the following serotonin 1A antibodies: Millipore-Chemicon, AB15350; Abcam, ab44635; Santa Cruz, sc1459 (goat) and sc10801 (rabbit). I didn’t try 24504 from Acris. Is it specific, Luis? Is its specificity strictly validated? Since all the antibodies I have tried gave different results, I’m really concerned about their specificity. Maybe all of them are not specific as Michele said.

I always used the Abcam one with good results. Still it depends. Are you incubating at RT or 4 degrees for 2 days. Depending on what I wanted to see, sometimes I had to incubate at 4 degrees. Lower signal but better marking.
