These symptoms appeard on the top leaves or new young leaves.

Did you also notice any insect there or curling of leaves?
However in order to confirm the presence of virus, extract the DNA from leaves and run the PCR with primers specifice to virus which you can find in the literature. If you get band of desired size, you can confirm it with sequencing as well. At the moment it is hard to say whether it is virus or nutrient deficeincy.
@Adnan: It isn't that easy. There are a lot of viruses occuring in cucumber. Should Rong Zeng buy primers (for different viruses) and perform x-PCR-reactions? And you need positive material... And maybe there is no PCR-cycler in the lab and all the required material... Think about it! Cheers, Nadine
Dear Nadine, Thank you for reply, Yeah It is not that easy and simple ,But I was just telling the ways to detecct the viruses, She can check for cucumber mosaic virus only, which is common in cucumbers. If I am wrong now, Please correct me as well. It is always intersting to learn from the experience which you have.

Hi, maybe it's not the cucumber mosaic virus. There is another way in detecting viruses: via ELISA.

I am Zeng Rong' s leader, Fu-Ming DAI. We have done some work on the unknown cucumber viral disease. In the field , only a few, not all the plants showed this kind of symptoms. We think it belongs to plant viral disease, not nutrient deficeincy . We also detected the samples via ELISA with the cucurbit crop common virus diagnostic kits (about 10 types). We hope to narrow the scope of virus species and get your suggestion for suspected virus species . Or else, we need more time doing this work.
Have you tried any biological characterization of this virus? For example is it mechanically or graft transmitted?

Thanks for all your help. As Dr. Dai said, All samples used various commercial ELISA kits to detect virus species, including CMV, ZYMV,WMV-1, WMV-2,SqMV, PVX,PVY, TMV, TuMV ELISA Kits, but no positive reaction was observed. We were not sure if some pseudo-negative in it. Maybe，we need do more work on PCR or RT-PCR to test the plants. We have no idea the way of transmission of disease. Test by mechanical inoculation or graft inoculation, really good advices for us!
The symptoms remind me of Cucurbit yellow stunting disorder virus (CYSDV). You might also check for a begomovirus, although the symptoms are not typical for this. They are diverse and can cause a wide range of symptoms. There are good general primers for begomoviruses.
RNA seq will tell you which virus it is, but this is not simple and it is expensive.
It may be a virus disease transmitted by aphids.try with spray of neem based insecticides like nimbicidine 30ml in 16 litres of water(1500ppm azadiractin )..pl. Reply if it works well.
Check Tospovirus and Thrips. Hot condition is suitable for disease and insect.
It looks like magnesium deficiency. What is the distribution in the field? Is it patchy or more or less uniform to all plants? To rule out CMV, you may try CMV immunostrips from AGDIA.
@Rong, I believe that I am a little bit late, but anyways. As all commenters said above, there are several reasons to have those symptoms. Among those reasons:
1. Viral infection (could be several viruses, not only CMV)
2. Nutrition problem.
3. Environmental issue (are those plants growing in greenhouse or plastic house?) it could be high humidity causing this problem, I have experienced that, and also high temperature could cause similar symptoms.
To troubleshoot this problem you can check the following:
1. the environmental conditions around plants (which could be the reason since the symptoms appeared lately. I am not sure but I could see the old parts of the plant were just healthy, is that right?) so maybe some changes in the environmental conditions happened recently.
2. if that is not the case, you can check for the nutrition issues, which can be done by having your record about the amounts of nutrients added to plants. check your record and if everything goes well, go to the next step.
3. Check for viral infection.(The reason I mentioned this point as the last one is because I don't see stunting or curling on top of plants, so it is most likely to be one of the two reasons above). The easiest way to do so is to buy Flaskit (immunostrips) which is a field test. It is very easy to use and fast. several companies produce them such as Bioreba and Agdia.

So many suggestions might be baffling you. As i see the pic looks the crop is grown under polynet house or protected structure. The whitefly is a serious pest in thesestructures.
Please check if you see any whitefly there. Im sure this is a cucumber crop infected with begomovirus. Only way to check is either collect fly from infected plantand leave on new healthy seelings. sametime you can rub the sap on 2-3 leaf stage seedlings.
Check for symptoms.
