I am looking for vectors containing GFP and RFP markers for transient expression in Arabidopsis protoplast (possibly also tobacco). Any suggestions? The constructs will be used for subcellular localization of two transporters; one which we believe is located in the inner envelope membrane of the chloroplast and the other in the thylakoid membrane. Thus, the reporter should be on the C-terminus for proper transport to the chloroplast. Readymade constructs with known localization in the chloroplast for co-localization would also be appreciated.
I have a number of plasmids containing hybrid coding regions where YFP, GFP or RFP are placed at the C-terminus of a protein...with restriction sites left inbetween to swap for your favourite gene (probably requires PCR amplification to make compatible sites, or simply PCR assembkly without sites.).
