In xylem, hydraulic conductivity measurements, what is the desired concentration of bleach that should be used in order to constrain bacterial growth on the samples before the vacuum process?

It is a big problem in the case. According to what we did. Ultrasonic anti bacterial technique should be a good solution for it.
I use a 5% bleach solution over night to bleach all tubing in the system (upstream of the plant segments) once a week. Rinse well afterwards. Other than that, I don't bleach anything.

In our research group we desinfect the destilled, filtered and degassed water for the XYL'EM apparatus with “Micropur Forte“ (MF 1000F, Kathadyn Produkts Inc., Wallisellen, CH, http://www.katadyn.com) with a concentration of 0,005% (v/v). Probably such desinfected water (silver ions combined with chlorine) would also be helpful for storing samples.
