In ovo injection of nutrients or antigens (as the case may be) has to be ensured to be delivered into the amnion of the embryo to have maximum result. To do this, the use of an commercial egg injector may be essential, especially if you are doing this commercially. If it`s just for research with small number of eggs, then you can adopt a manual (hand) injection method.
Specific time of injection is also something to keep in mind as you want to target around when the developing embryo`s amnion is at it`s highest volume to increase your chance of amniotic delivery (for broilers, 17 to 18.5 day of incubation).
The specific type, form, and amount of nutrient you will be injecting is also of importance. Toxicity is something you want to have in mind

First, ensure that the eggs are fertilized (e.g. by candling), then prior to in ovo injection clean the eggs with 70% alcohol (avoid contamination during injection), then use an egg puncture to make small holes on the eggs (at the broad end of egg), then using a syringe (e.g 1mL x 2.5 cm) with 23-gauge needle you can slowly inject the nutrient(s) into the amnion of the eggs. This will do for an experimental setup.
