Hi Agustin, since the his tag is located at the c-terminal side, removal of the his tag using TEV will leave behind six amino acids (from the TEV recognition site). I am looking for proteases that can completely remove his tag and its recognition site.
Bovine carboxypeptidase A from Sigma will remove polyhistidine tags from the C-termini of recombinant proteins. I don't think this is widely known, but BCPA is a glycoprotein, so it can be removed from the digest by capturing it on an immobilized lectin column (e.g., concanavalin A).
