I want to determine the solubility of Clofazimine and then to try the enhancement of its solubility using various lipid solvents, lipid-water mixtures and buffers.
You can use Vitamin E TPGS. This water soluble derivative of Vitamin E is reported to enhance the solubility of lipophilic drugs by manifolds. Cyclodextrin inclusion complexes can be useful.
Dear Dr. Jan, I saw the question that you posted back in 2012 about the enhancement of Clofazimine solubility. I have tried to improve its solubility without any success. I tried to do it using Vitamin E TPGS by mixing dried E TPGS and Clofazimine, heating up the mixture at 45oC, placing the warmed mixture at 4oC for solidification, and then scratching 10 mg to re-suspend it in 1 ml of water. But, I still see Clofazimine precipitants in the tube. I am wondering if you or someone else could come up with a successful protocol either using vitamin E TPGS or something else that would allow a total resuspension of Clofazimine. Resuspending Clofazimine in lipids would not help me for my research purposes. Thanks a lot! Rossio
