I am going to grow C. scindens and C. sporogenes. We have the air-tight boxes where you put an anaerobic pouch, close the box, and put it in the incubator, but we are not equipped with an anaerobic chamber to handle the bacteria under anaerobic conditions. Is it possible for me to handle the bacteria under aerobic conditions (less than an hour under aerobic conditions) and incubate under anaerobic?
Not at all recommended. However, you can give it a chance in nitrogen or CO2 rich environment.
Some clostridia are strict anaerobes and will not grow if exposed to oxygen, even during a brief period of time. Others, such a C. perfringens, are more aerotolerant and will grow if manipulated under aerobic conditions (but they still need anaerobic conditions to grow). If you do attempt manipulating your isolates under aerobic conditions, keep exposure to oxygen at the shortest possible. Before attempting this, I would advise checking if the species you mention are very strict regarding anaerobiosis.
