I found many papers that claimed that AB1-42 1uM is not enough to significantly increase Caspase 3 activity and significantly reduce Bcl-2 expression in SH-SY5Y cells but I also found some papers finding that AB1-42 1uM can increase the expression of caspase 3 and reduce the expression of Bcl-2. At present, I do a western blot to investigate whether AB1-42 1uM can increase caspase 3 expression and reduce Bcl-2 expression in SH-SY5Y cells, and the result is not so stable that I suspect whether it is because of my performance or the natural of this substance. the toxicity of the AB42 peptide depends on the form of preparation you need to prepare the powder in DMSO and after diluting it in PBS pH7,4 and incubating it at 4 Celsius degrees overnight to form oligomers, Abeta 1-42 solution 1uM, 5uM and 10 uM will induce cell death and apoptotic changes. I may forget some details, please check in some of my papers with cRaf inhibitors in vitro. Try primary cultures is a better model than cell lines
