Hi. I am doing a in vivo study using cadual artery injection for bone metastasis. I am using PC3REDGFPFLUC cells. I have question regarding bone metastasis spreading.

During the injections, half of the injections went well. I could easily inject on the first or second time in the cadual artery. I didnt feel any resistance during the injections. It was easy to inject, I could see a flash of white in the tail and also blood came out from the artery after I withdrawn the needle.

The thing is now (two weeks after injections), when I use D-luciferin to image the spread of PC3REDGFPFLUC cells, I only see signal from the tail where the injections probably occured and no spread to bones or anything else. What could be the problem here?

Could it be that I used to many cells for the cell suspension so it got stuck in the artery? I used 1 000 000 cells in 100 ul PBS and I pipetted the cell suspension a few times before each injection. I am grateful for suggestions.
