I am working on macrophage polarization to M1 and M2.
I have noticed that most of publications use Bone marrow derived macrophages from mice over Macrophages derived from PBMC.
Is there any particular reason why BMDM is preferred over PBMC derived Macrophages?
When choosing between BMDMs and PBMCs, researchers often prefer BMDMs due to their tissue-specific origin, purity, and functional characteristics. BMDMs closely resemble tissue-resident macrophages and allow for more controlled studies. PBMC-derived macrophages, while versatile, represent a broader immune response and are less specific to a particular tissue context. The choice depends on the research question and experimental design.
Another reason is that, that you would have to pool various mice (8-9) to isolate monocytes from peripheral blood and reach a reasonable cell number. From bone marrow you harvest and differentiate about a few million (20-40) per mice. Thereby, you would obtain enough cells from one mice for various conditions.
