I am hoping to obtain some guidance regarding the best method to store mushroom fruiting bodies from the field for DNA preservation and subsequent Illumina sequencing.

From what I have read already my main options are:

1) Drying - recommended below 60 degrees.

2) Immediate freezing - I have seen this range from immediate freezing at - 80, to -4.

3) Storage in ethanol.

I am leaning towards drying as a preservation method. Does anyone have any advice for/against any of these methods? There are a number of papers that explore various drying temperatures etc, but I am yet to see a comparison between storing in liquid buffer versus drying. I wish to barcode the ITS region for simple fungal identification, followed by CO1 sequencing to explore the associated invertebrate community.

Thank you in advance,

Kirsty.
