I am working on a bacterial P450 enzyme. I am wondering whether anyone knows of a P450-reductase that can be purchased or the general way to proceed to see activity. I have the enzyme and the substrate but not sure on what type of reductase to use (ferredoxin, cytochrome b?).
Interesting question. If is a self-sufficient enzyme, you can find interesting this paper: "A Self-sufficient Cytochrome P450 with a Primary Structural Organization That Includes a Flavin Domain and a [2Fe-2S] Redox Center", by Roberts et al. (is a little bit old, I know)
That is pretty cool, but based on homology mine doesn't have a flavin domain nor an Fe/S domain.
Mmm, I'm quite sure you will find this very useful for preliminary studies: "Cytochrome P450 systems—biological variations of electron transport chains", by Hannemann et al. And I suggest you also this review "A selective review of bacterial forms of cytochrome P450 enzymes", by Lewis and Wiseman, very useful for bacterial P450
More curious from a practical point of view. Anyway, figured out so when in doubt use ferredoxin and ferredoxin reductase from sigma. Mammalian P450's have their own reductases which you can also buy.
