I need to analyze total carbohydrates in different rice varieties but due to some reasons I am unable to import Megazyme available carbohydrates kit. Can anyone recommend other methods/kits of available carbohydrates detection
There are alternative methods and kits for detecting total carbohydrates in rice varieties. Here are some of them widely used methods:
Phenol-Sulfuric Acid Method:
This method involves treating carbohydrates with concentrated sulfuric acid and then reacting with phenol. The resulting color change can be quantified spectrophotometrically. This method is commonly used for quantifying total carbohydrates, including simple sugars.
Anthrone Method:
In the Anthrone method, carbohydrates are hydrolyzed and then treated with anthrone reagent. The resulting complex absorbs light at a specific wavelength, allowing for quantification. This method is often used for total carbohydrate analysis.
DNS Method (3,5-Dinitrosalicylic Acid):The DNS method is commonly used for reducing sugar quantification. It involves reacting reducing sugars with DNS reagent under acidic conditions, resulting in a color change. This method is particularly suitable for samples with reducing sugars.
Bial's or Orcinol Method:
Bial's or Orcinol method involves the hydrolysis of carbohydrates followed by reaction with orcinol reagent. The resulting complex can be quantified spectrophotometrically. This method is suitable for the analysis of a wide range of carbohydrates.
