Good day! How we are having a project, in which one of the stages consist of evaluation of the structure of hemicellulose polysaccharides. However, not so many literature is dedicated to such a studies. In some of them, water solutions of hemicelluloses are used. However, its individual macromolecules tend not to fully hydrate, thus supramolecular aggregated chains remain present even in very diluted solutions. Also, different solvents may be used (buthanol), different types of pre-treatment (ultrasonication) or even hemicellulose dissolving with Lithium salts solution. Thus, I wanted to ask are there any other possibilities to visualize hemicelluloses with AFM to obtain it in single-molecule state (as on Figure)? Many thanks for any kind of suggestions or articles.
