My lab does a lot of gel extraction for cloning purposes. We fly through the columns in the Qiagen extraction kit, but we still have significant amounts of buffer left. Are there other columns we can use to keep costs down since it is really expensive to keep buying new kits with reagents we don't need? We do not want to use silica or other beads, just columns.
If you are using it only for gel extraction of DNA just use the freeze and squeeze method as long as you dont use Boric acid in ur running buffer.
THis process does not require any kits .
Step1 cut out gel piece of interest.
Place it in a 0.5 tube and freeze it.
Step 2 Pierce the bottom of the tube while frozen with a small gauge needle.
Step 3 place the first tube in a 1.5 ml tube and thaw at room temp and spin at 8K G for 2 min.
The eluate contains your DNA.
