Is there are linear relationship between direct count and standard plate count of bacteria?

Yes. There is a direct relationship between the ODS (optical density) at 660 nm and the standard plate count. But you will have to plot both curves since the type of bacteria (grapes vs. chains) influences the slope of each curve.
