I would like to run LC-MS/MS with a 300um column, but the commercially available ones are only 15 cm. There are many papers that discuss packing 50-150um columns in 365um OD capillary, but has anyone come across a similar paper for packing 300um ID columns?
We packed 200 um monodisperse Polystyrene Copolymers in long fairly large diameter columns. Column packing is art with many variables!! (Particle physics, Particle chemistry, hydrodynamics, Column parameters and Packing method parameters) Once you have established the parameters you have to design a method and pack it say 3 times to measure reproducibility). As I said it is art. Thus you have to develop your own skills. Alternatively, give it to people that pack columns every day and use their skills against a fee.
