After culturing feacal samples using MacConkay agar. The Indole test requires tryptophan for identification of the bacteria E. Coli. I have no tryptophan. Is there another alternative way/ways to specifically identify the bacteria.

By performing Indole test alone you cannot say that strain is E.coli. There is a list of other biochemical tests (IMVIC) by which you can confirm.
