RNA from Mentha plant medical leaves was extracted using IHBT Modified method. According to the electrophoresis gel attached photo, Are these bands compatible with numbers that show spectrum for concentration and quality? Because the 260/230 ratio and the quantity is low. If you don't give the volume loaded for each sample there is no way to answer the question... What is obvious is that lane 2 contains far more than 3 lanes and 4 which are equivalent...
the od260/280 levels are good for dna but low values for the od260/230 suggest that there is some contamination. Possible contaminants apart from residual DNA are
- Carbohydrate carryover (often a problem with plants).
- Residual phenol from nucleic acid extraction.
- Residual guanidine (often used in column based kits).
- Glycogen used for precipitation
- If there are contaminants it is hard to say if the samples are directly comparable which might be the case in sample 1 where there may be high molecular weight dna near the well which would have contributed to the OD260
If you have loaded the same volume of each sample, there is definitely an issue as the sample giving the most intense pattern is the one exhibiting the lowest concentration...

For this plant TRIzol cannot have a good respond, because of phenolic and polysaccharide compounds, I have to use a manual method.
