Hi everyone,

Recently, I stained( immunofluorescence staining) eyeball sections with APC conjugated antibody. I could observe many positive cell under confocal microscope. But there were extremely lightly stained. Even I increased the concentration of antibody for 4 times and prolong the incubation time from 1h to 3h.  The result was almost same.

So, I wonder if these extremely lightly stained cells are actually positive cells? How to improve?

Thanks,

Jingliang
