I'm trying to build a cosmid for B. subtilis, but there are no commercially available cosmids with a gram-positive ori already installed. I was thinking of obtaining a commercial cosmid (SuperCos 1) for E. coli and cloning a gram-positive ori into the vector so I can use that cosmid's MCS to insert my desired gene clusters. Would this be a feasible strategy, or would it be easier to start with a plasmid with a gram-positive ori and install the cos site? Any advice or resources would be appreciated. Thanks.
