The mutant seeds may not germinate because of mutations in genes essential for seed germination, such as those involved in GA signaling. As the dose of mutagen/mutation increases, the disruption to these genes is more severe, resulting in a lower germination percentage.
You can try a few things:
Stratify the seeds at 4°C for several days before planting. This can help to break dormancy and improve germination.
Use fresh seeds for mutation treatment. Because Arabidopsis seeds have a relatively short shelf life.
Use a sterile growing medium to avoid fungal contamination.
Plant the seeds at the appropriate depth. Arabidopsis seeds should be planted shallowly, just below the surface of the growing medium.
