You are on the right track. It's important to note that this kit is designed to precipitate (well capture in the correct terms) chromatins. hence the name, Chromatin immunoprecipitation ChIP. You will end up with your DNA+Chromatine+ DNA-binding protein of interest that requires further purification perhaps- really depending on your aim to what to do with it. I would suggest to be very aware of the salt concentrations you are using as some attached proteins may dissociate with a moderate concentration of salt. Unfortunately, there is no magic concentration that I can suggest (it highly varries between proteins- Histones are strongly attached to DNA for example) but try to find more info about your protein in the literature or even sequence analysis finding it's isoelectric point etc.

Thanks very much, it's a practice essay question I'm working on and it's undergrad level. We haven't covered this in lectures yet but I've found my best bet at learning something is to do it this way and jump in (so that's why it probably sounds like a really basic question).

The question asks how we would purify a Dna binding protein and then study the Dna binding activity of the isolated protein. I guess they wouldn't expect us to come up with some novel method and are looking for the most common methods, what kind of thing should I be looking at as to how to study them in isolation? A PAGE variant? Thanks for all the advice

I can lead you into the way and you can find the answer yourself, but yeah the first step is to use immunoprecipitation. If you have an antibody specific to that protein you will be able to isolate the protein and the DNA attached to it... if you need to study the protein itself you might need to go about separating the DNA from the protein (usually by increasing salt and running on a column-- try to find out the type of the column you can use- and then yeah doing PAGE- etc) or to study the DNA that binds specifically to the protein you need to digest the DNA without implicating the protein then again separate the DNA from the protein but now study the sequence of the DNA that the protein was originally attached to... the DNA digestion step will digest everything except the bits that your protein is attached to.... that's actually the basis of studying epigenetics and many other fields regarding DNA binding proteins... think about that and try to Google ChIP then go into images and you will have a clearer picture :) good luck
